There were 26 questions found in this category:
  1. questionCan ethanol be used on RC Chambers?
Yes, they are made from polycarbonate and should be resistant to EtOH.

  2. questionCan the RC-21B/RC-21BR be used on a confocal microscope?
Yes, the RC-21B and RC-21BR can be used on a confocal microscope. In general, one critical consideration when using imaging and recording chambers in confocal imaging is the small working distance of the objective. All Warner chambers use a #1 coverslip as the chamber floor which should allow vi ...

  3. questionCan the RC-29 (open bath chamber for slice studies with atmospherics) be used with cultured cells?
Yes. Use of the RC-29 open bath chamber with atmospherics is not restricted to slice studies. Simply replace the slice anchor with a 24x50mm glass coverglass with cultured cells. Vacuum grease is used to seal the coverglass to the chamber while the platform holds them both together.

  4. questionCan the TSP wash remove vacuum grease from plastics?
The short answer is that the TSP wash procedure will not remove vacuum grease from plastic. Your best option is to remove the grease by mechanical means (e.g., rub it off with a Kimwipe). You should then be able to clean things up using the TSP wash.

  5. questionCan Warner Series 20 Perfusion Chambers be used with #0 thickness Cover Glass
Nothing speaks against the use of #0 thickness glass coverslips with our Series 20 chambers. However, you might want to use a P- or PH- platform with screw-clamps rather than a PM- platform with magnetic clamps. The screw- clamps will allow you to adjust the clamp pressure. Be careful not to br ...

  6. questionHow can a ground wire be attached to a chamber that doesn't have an agar bridge?
A silver-silver chloride ground electrode can be placed directly into the suction well downstream from the sample. Be aware, however, that if you do not continuously perfuse the sample, the silver wire may be able to contaminate the bath.

  7. questionHow can a steel Sutter stand be insulated from a heated series 20 chamber?
When heating a sample in a chamber an insulator can be used to prevent the steel Sutter stand from acting as a heat sink. The insulator can be made out of a thin layer of plastic, cardboard, or even a latex glove.

  8. questionHow can I generate and maintain a hypoxic solution for delivery to a chamber?
This is normally achieved by bubbling an O2-free gas through the solution until the O2 load has been depleted. Keep in mind, however, that PE, Tygon, and Teflon tubing are all gas permeable, and therefore will expose your perfusing solutions to O2 during transit. One option to address this is to ...

  9. questionHow can the development of air bubbles in an open bath perfusion chamber be eliminated?
The amount of gas a solution can hold is dependent on the solution temperature. As a result, heating a solution, usually just before delivery to the chamber, can result in outgassing and the appearance of bubbles in the bath.A simple way to address this issue is to preheat your solution, in the ...

  10. questionHow do I clean the Warner chambers?
We recommend using our TSP wash. A reference to this procedure can be found below

  11. questionHow do I increase the stability of my bath height when using an open chamber?
If the chamber bath height is not stable it is usually due to the formation of a meniscus at the aspiration tube. This can lead to an uneven aspiration rate. There are two simple ways to improve bath height stability.1. Use a strong suction on the aspiration end.2. Put a small piece of Kimwipe in ...

  12. questionIs it possible to use temperature feedback from the bath to regulate an inline solution heater?
Unfortunately, no. Feedback in inline solution heaters is derived from thermistors located within the heater body. This prevents oscillations in temperature which can arise when the temperature sensor is distal to the heat source..

  13. questionIs there a chamber that can be used to generate a chemical gradient while also perfusing a sample?
We are not aware of a system that has been developed whereby a sample can be perfused while maintaining a well defined gradient. Every method that comes to mind would result in either a poorly defined gradient or a plus/minus chemical step, which is all or nothing. However, Ibidi sells a chamber ...

  14. questionIs there a chamber that can perform electrophysiology under hypoxic conditions?
There are several ready-to-use options for doing electrophysiology under hypoxic conditions. To generate hypoxic conditions with each of the following chambers simply pump in a gas such as nitrogen or argon (preferably argon as it is heavier than air). The positive pressure of the introduced gas ...

  15. questionWhat cables are needed to connect al DS3 isolated stimulator and a DG2A pulse generator to a Series 20 imaging chamber?
A BNC to BNC cable (10 ft long, order# 4607012) and a cable with 1 mm female jacks to 2 mm pins on 0.75" centers (also 10 ft long, order # 99-0643) will connect the devices.

  16. questionWhat chambers do the Sutter chamber columns accommodate?
The Sutter MT-150/W2 chamber column accommodates any Series 20 chamber and platform. You must inform Sutter if you intend to use a Warner PM series platform as they will supply a different adapter. The MT-150/CUST is a custom built Sutter chamber column that is designed to accommodate a user speci ...

  17. questionWhat is the working volume of the RC-26GLP?
The RC-26GLP open bath chamber has a volume of 234 u l (0.234 ml) per mm of solution height above the base coverslip. The horizontal outline of this chamber is identical to the RC-26G. However, the total height of the bath in RC-26GLP is 2 mm (as compared to 4 mm for the RC-26G). The shallow bat ...

  18. questionWhat length of electrode is exposed to the bath in your field stimulation chambers?
    Chamber Exposed Electrode (on each side of bath)RC-21BRFS 12mmRC-27NE 25mmRC-37FS 19mmRC-37WS 13mmRC-47FSLP 15mm

  19. questionWhat type of grease is used for sealing the gaskets and coverglass on RC Series chambers?
The grease we recommend is the Dow 111 silicone grease that is normally used with vacuum pump tubing line connections. It is also used to seal glass dessicators. We sell two products that contain a tube of this grease. Order number 64-0275 is simply a tube of Dow 111 silicone, while order number ...

  20. questionWhen is in-line heating preferred over chamber heating?
Generally speaking, heating a platform alone is a good strategy only for very slow or non-flow conditions. The solution loses most of its heat enroute to the chamber. Under the more common flow conditions of 2-10 ml/min, an inline heater is a better way to deliver heated solution to a chamber.Co ...
